Abstract
Background: Hypothyroidism is One of the most frequent chronic disorders worldwide, which occurs in the cases of decrease in the thyroid hormone secretion less than the normal value. Thyroid hormones play an important role in keeping the normality of salivary gland (function and histology) as it is crucial for homeostasis of oral health. The aim of the study: Examine the role of Ashwagandha Roots extract versus thyroxine hormone to attenuate the histological effects of hypothyroidism in rats after postnatal exposure to Propylthiouracil. Materials and Methods: Ten healthy pregnant albino rats were procured. Each pregnant rat was housed separately in clean rodent plastic until the newborn pups were noticed. At PND3 forty male pups ( albino) were randomly categorized into 2 main groups ,group A as a control group , and group B which was given (1mg/kg) Propylthiouracil (PTU) orally, for three weeks. At PND22, group B divided into 3 subgroups: group B1, the hypothyroid group that remains without treatment; group B2 hypothyroid group that received an aqueous extract of Ashwagandha Roots (200 mg/kg) for 21 days; and group B3 that received Levothyroxine (4µg/100g/day) also for 21 days. The submandibular gland specimens were processed for histological techniques at the end of the experiment for all groups . Result: the hypothyroid group’s submandibular section showed atrophy of acini with degeneration of epithelial cells lining it, necrosis of epithelial cells lining granular convoluted tubules and degeneration of epithelial cells lining striated ducts. While administration of Ashwagandha Roots extract showed improvement in the most of these histological changes.
