Abstract
Imidacloprid (IMI) is an important insecticide that has been widely applied to control insect in agricultural fields. Aim of the study: to evaluate the toxicity risk of Imidacloprid insecticide on mice males and the possible protective effect of vitamin C.
Materials and methods: The present study was carried out on sixty albino mice. They are divided equally into three groups, each of twenty mice. The first group served for control and divided into two subgroups, one receiving corn oil and the second as positive control receiving cyclophosphamide. The second group divided into two subgroups, one received small dose (1/25 of LD50) of IMI and the second received the small dose with vitamin C 10mg/kg orally gavaged. the third group divided into two subgroups, one received big dose (1/10 of LD50) of IMI and the second received the big dose with vitamin C 10mg/kg orally gavaged. Specimens from mice testis were taken after sacrificing of animals for histological, cytological and molecular techniques and blood samples were collected for biochemical assessment.
Results: Histological examination findings revealed distinguishable influence of big dose of IMI as maturation arrest of seminiferous tubules at the level of spermatocytes. By contrast of protected big dose of IMI with vitamin C showing normal spermatogenesis of seminiferous tubules to the level of spermatozoa .Sperm examination of treated Mice with big IMI dose showed different abnormalities like head loss, bent or coiled tail with multiple abnormal sperm structures.Laboratory findings revealed that treated mice with big dose reflected testosterone decreasing level. RAPD fingerprinting technique was used to detect and track GTS percentage, big dose of IMI showed decreased genome stability with 20 %.
Conclusion: Distinct toxic effects of IMI insecticides were indicated. Also vitamin C administration with IMI revealed some protective effects for mice exposed.
