Role of Myokine Decorin and Cholecalciferol as Therapeutic Targets in Polycystic Ovary Syndrome induced by estradiol valerate in a rat model.

Abstract

Background: Polycystic ovary syndrome (PCOS) causes psychological, reproductive and endocrinological disorders. Decorin (DCN) affects folliculogenesis and ovulation.Vitamin D receptors were detected in ovaries, DCN and vitamin D3 (Cholecalciferol) may accompany ovarian function.This study assessed the therapeutic option of DCN and vitD3 on ovarian function in PCOS provoked by estradiol valerate(EV) in rat.

Methods:Sixty adult rats (4groups), Group Ι (control):24 rats divided into 3 equal subgroups. Subgroup Ia: Rats received single intramuscular(IM) injection of saline, then after 60 days, they were given saline by intraperitoneal (IP) every other day and saline orally per day by gavage for 21days.Subgroup Ib: received single IM injection of saline, then after 60 days, rats were given Cholecalciferol orally by gavage daily for 21 days. Subgroup Ic: received single IM injection of saline, then after 60 days, rats received DCN IP every other day for 21days.GroupII(PCOS-induced group):12 rats received single IM injection of EV, then after 60 days treated as subgroup Ia.Group III (PCOS treated by vit D):12 rats received single IM injection of EV, then after 60 days, treated as subgroup Ib.GroupIV (PCOS treated by DCN):12 rats received single IM injection of EV, then after 60 days, rats received IP DCN every other day for 21 days.At end of this study, assessment of serum insulin, sex hormones and glucose was done. Ovarian tissue oxidative and inflammatory markers, histopathology, transforming growth factor-β1(TGF-β1) and androgen receptor(AR) immunohistochemistry were assessed.

Results: DCN and vitD improved inflammatory and oxidative stress markers, hormonal profile, ovarian structure, increased TGF-β1 and decreased AR immunoreactivity.The effects of DCN were better than vit D.

Conclusion: DCN and cholecalciferol can be used as therapeutic agents for PCOS with preference for DCN.
