The Possible Protective Effect of Dapagliflozin and Moringa Alone or in Combination on The Liver of Type 2 Diabetic Albino Rats: A Biochemichal, Histological and Immunohistochemical Study

Abstract

ABSTRACT:
Background: Type 2 diabetes mellitus (T2DM) has injurious impact on liver due to high levels of blood sugar, fats, oxidative stress, and an increased incidence of inflammation. These factors can consequently lead to various liver disorders.
Aim of the work: To ascertain impact of T2DM on histological structure of liver molecular and metabolic dysfunction and the potential protective function of dapagliflozin and moringa
Materials and Methods:
Fifty adult male albino rats were separated into five groups: control group, non-treated diabetic group, diabetic treated with dapagliflozin group, diabetic treated withMoringa oleifera extract group and diabetic treated with dapagliflozin plus Moringa oleifera extract group. After 4 weeks, blood samples were took via cardiac puncture for biochemical parameters evaluation expression of miRNA 34a, Secretagogin, sirtulin genes also liver samples were processed for histological and immunohistochemical studies.
Results: Diabetes mellitus induced impairment on liver function with disturbed liver architecture. Administration of Dapagliflozin and Moringa alone in group III & IV revealed non-significant decrease in mirRNA34a and secretagogin expression with moderate improvement in biochemical and histological finding. Otherwise, after treatment of diabetic rats by Dapagliflozin plus Moringa in group V, mirRNA34a and secretagogin expression levels reduced significantly and sirtulin elevated compared to non- treated diabetic rats. The improvement shown in liver function indicators, oxidative stress markers and lipid, glucose levels. There is also an improvement in histopathological findings in group V with a significant decrease in main area distribution of collagen fibers, positive caspase 3 cytoplasmic immunoreactivity and positive PCNA nuclear immunoreactivity in compared to untreated diabetic group.
Conclusion:
Administration of dapagliflozin plus moringa oleifera extract significantly ameliorated the hepatic alteration induced by diabetes mellitus type 2 in rats.
